1. GnRH releasing analogs
1.1. MOA: GnRH analogs initially stimulate then inhibit FSH and LH = decrease estrogen and testosterone
1.2. leuprolide; goserelin
1.3. for prostate and metastatic breast cancer
1.4. AE: hot flashes; decreased bone mass; libido decline
2. Antiestrogens
2.1. MOA: selective estrogen receptor (ER) modulators/downregulates expression
2.2. Tamoxifen; fulvestrant
2.3. ER positive breast cancer tx and prevention in high risk pts
2.4. AE: hot flashes; n/v; monitor for endometrial cancer
3. antiandrogens
3.1. useful in prostate cancer
4. Aromatase inhibitors
4.1. MOA: inhibit aromatase = less estrogen
4.2. Anastrozole; Letrozole
4.3. for Breast cancer
4.4. AE: well tolerated
